What are sleep supplements?
Sleep supplements are products designed to be used as part of an evening or bedtime routine.

A human study involving people who were unhappy with their sleep used 3g of glycine before bedtime.

Why is magnesium used in sleep supplements?
Magnesium is an essential mineral.
Your body needs it for many normal processes, including normal nervous system function and normal muscle function.

A systematic review looked at 9 studies containing 7,582 people.

What is magnesium bisglycinate?
Magnesium bisglycinate is a form of magnesium where magnesium is bound to glycine.

Has magnesium bisglycinate been studied for sleep?
Yes.
A 2025 randomised, double-blind, placebo-controlled study included 155 adults aged 18 to 65 who reported poor sleep.
Participants took either magnesium bisglycinate providing 250mg of elemental magnesium each day or placebo for four weeks.
Insomnia Severity Index scores fell by 3.9 points in the magnesium group compared with 2.3 points with placebo.
The between-group result reached statistical significance (p = 0.049), although the researchers described the effect as modest, with an effect size of Cohen's d = 0.2. (PubMed PMID 40918053)

What is L-theanine?
L-theanine is an amino acid found naturally in tea.

What is L-tryptophan?
L-tryptophan is an essential amino acid.

Has tryptophan been studied for sleep onset?
Yes, although much of the direct human sleep research is older.
One controlled study tested 250mg, 500mg and 1g of L-tryptophan against placebo in 15 people who reported taking more than 30 minutes to fall asleep.
The 1g dose significantly reduced sleep latency. (PubMed PMID 469515)
A review of around 40 controlled studies concluded that doses of 1g or more most consistently increased subjective sleepiness and reduced sleep latency, particularly in people with mild sleep-onset problems. (PubMed PMID 6764927)

What is valerian root?
Valerian is a plant with the botanical name Valeriana officinalis.
Its root has a long history of traditional use in products aimed at relaxation and sleep.
It is also one of the more heavily researched herbal sleep ingredients.
What does the valerian research say?
The answer is mixed.
A 2020 systematic review included 60 studies and 6,894 participants.
The researchers found that results varied between products and preparations, with extract quality likely helping explain some of the differences. (PubMed PMID 33086877)
A newer 2024 umbrella review reached a cautious conclusion: evidence for valerian treating insomnia itself remained weak or inconclusive, although subjective sleep quality may improve. (PubMed PMID 38359657)
Valerian is a good example of why "clinically studied ingredient" does not automatically mean every trial has been positive.

What is lemon balm?
Lemon balm is a herb with the botanical name Melissa officinalis.
It has traditionally been used in calming preparations and is now being studied in modern sleep research too.
A double-blind placebo-controlled crossover study of a standardised lemon balm preparation reported an average Insomnia Severity Index score of 6.8 during treatment compared with 9.7 with placebo.
That was a difference of 2.9 points (p = 0.003).
Researchers also reported that 87% of participants said their sleep improved compared with 30% during placebo. (PubMed PMID 39683592)
However, this was a particular standardised lemon balm formulation, so the exact results should not automatically be applied to every lemon balm extract.

What is chamomile?
Chamomile is one of the best-known herbs associated with bedtime.

It has also been studied clinically, although the evidence is not completely consistent.
A placebo-controlled pilot study involving 34 adults with chronic insomnia tested 270mg chamomile extract twice daily for 28 days.

Other research has been more positive. A trial in 60 older adults used 200mg chamomile extract twice daily for 28 days and reported improvements in sleep quality. (PubMed PMID 29154054)
A 2024 systematic review and meta-analysis has since brought together the available clinical trial evidence. (PubMed PMID 39106912)
Chamomile is a traditional night-time botanical with human research behind it, but it would be misleading to describe the evidence as uniformly positive.

What do sleep supplement patents tell us?
Patents are not proof that a supplement works.
They can, however, show how researchers and product developers group ingredients according to different parts of the sleep process.

Sleep and recovery are closely linked.
While you sleep, your body is not simply "switched off".
